Carbohydrate Antigen 19-5 (CA19-5) Elisa Kit uses competitive enzyme-linked immune detection method to determine the level of test substance in the specimen. The monoclonal antibody of carbohydrate antigen 19-5 (CA19-5) is coated on a microplate to prepare a solid phase carrier, and the biotin-labeled antigen and the antigen to be tested (standard or sample) are simultaneously added to the microwell of the coated antibody, the antigen to be tested and the biotin-labeled antigen compete for binding to specific antibodies. After incubation, the unconjugated material was removed by washing, then HRP-labeled avidin was added, and after incubation and thorough washing, the substrate TMB was added for color development. TMB is converted to blue under the catalysis of peroxidase and converted to the final yellow color by the action of acid. The higher the concentration of the sample to be tested, the more the binding of the labeled antigen and the antibody is inhibited, and the lighter the coloration. The depth of color development is positively correlated with the amount of enzyme, but negatively correlated with the content of the substance to be tested in the sample. The absorbance (O.D. value) was measured at a wavelength of 450 nm using a enzyme-labelling measuring instrument to calculate the sample concentration.
